Capturing Pluripotency era
Gail R. Martin's 1981 work isolated mouse embryonic stem cell lines from blastocysts, establishing stable pluripotent cultures. Around the same period, Martin Evans and Matthew Kaufman independently derived mouse ES cell lines and laid down culture conditions that sustained long-term self-renewal. Leukemia inhibitory factor and embryoid body formation emerged as core tools for maintaining pluripotency and guiding multilineage differentiation in vitro. Promoter traps and integrated functional genomics linked gene activity to pluripotent culture, enabling forward screens and developmental dissection within these systems.
